ABSTRACT

Objective:To establish the high performance liquid chromatography (HPLC)fingerprints of Zhuanggu Guanjie Pills, which provided the basis for its quality evaluation.Methods:HPLC was used with Agilent Eclipse XDB-C 18 column (4.6 mm×250 mm, 5 μm);mobile phase was acetonitrile-0.1% acetic acid water; gradient elution; flow rate was 1 ml/min; column temperature was at 35 ℃; detection wavelength was 254 nm; injection volume was 10 μl. The HPLC fingerprints of 15 batches of Zhuanggu Guanjie Pills were established and similarity analysis was carried out, and the contents of 18 components were estimated. Results:In the fingerprint study, isopsoralen was used as the reference peak, 40 common peaks were marked and 18 peaks were identified and the similarity between the fingerprints of 15 batches of Zhuanggu Guanjie Pills and the control fingerprints was greater than 0.99.Conclusion:This method is easy to operate and has high accuracy, which can provide basis and reference for the quality evaluation of Zhuanggu Guanjie Pills.

ABSTRACT

This paper summarizes professor GUAN Ling's clinical experience in the treatment of knee osteoarthritis (KOA) with structure-based medical acupuncture (SMA). Based on anatomy and biomechanics and through accurate physical examination, SMA adjusts the mechanical imbalance of muscles to relieve KOA dysfunction, and releases nerve compression to attenuate pain symptoms of KOA. In reference to traditional acupoint selection, and in association with painful areas and mechanical deduction, ashi points located at the rectus femoris, vastus intermedius, vastus medialis and vastus lateralis muscles, etc. are specially stimulated with acupuncture; and the rehabilitation training and health education are the adjuvant treatment for the patients.

ABSTRACT

Objective:To identify the anti-acetylcholinesterase active ingredients in <italic>Aconitum tanguticum</italic>, so as to lay the foundation for finding new anti-Alzheimer's disease (AD) drugs. Method:The anti-acetylcholinesterase active fractions of <italic>A. tanguticum</italic> were screened by the modified Ellman's method, and the chemical composition of the active fraction was analyzed by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS). The chromatographic separation was performed on an ACQUITY UPLC BEH C<sub>18</sub> column (2.1 mm×50 mm, 1.7 μm) with acetonitrile (A)-0.4% ammonia aqueous solution (B) as mobile phase for gradient elution, and the column temperature was set at 30 ℃ with the flow rate of 0.4 mL·min<sup>-1</sup>. Phase A of the dichloromethane fraction changed with time as follows:0-3 min, 5%A; 3-7 min, 5%-20%A; 7-11.5 min, 20%-33%A; 11.5-15.5 min, 33%-50%A; 15.5-20.5 min, 50%-80%A; 20.5-23 min, 80%-85%A; 23-25 min, 85%-95%A. Phase A of the <italic>n</italic>-butanol fraction changed with time as follows:0-2 min, 5%A; 2-8 min, 5%-20%A; 8-11 min, 20%-33%A; 11-15 min, 33%-95%A. Mass spectrometry was performed on electrospray ionization, data were collected in positive ion mode, and the detection range was <italic>m</italic>/<italic>z</italic> 100-1 500. Result:Both the dichloromethane and <italic>n</italic>-butanol fractions had a certain inhibitory effect on acetylcholinesterase, their half inhibitory concentration (IC<sub>50</sub>) values were (64±4.4) mg·L<sup>-1</sup> and (85.7±3.8) mg·L<sup>-1</sup>, respectively. By UPLC-Q-TOF-MS/MS analysis, a total of 21 alkaloids were identified from the dichloromethane fraction, and 11 alkaloids were identified from <italic>n</italic>-butanol fraction. Guan-fu base Ⅰ, found in both fractions, was first discovered in <italic>A. tanguticum</italic>. Conclusion:Diterpene alkaloids are the main anti-acetylcholinesterase substances of <italic>A. tanguticum</italic>, which is worth further exploration.

BACKGROUND@#Prenatal stress can cause neurobiological and behavioral defects in offspring; environmental factors play a crucial role in regulating the development of brain and behavioral; this study was designed to test and verify whether an enriched environment can repair learning and memory impairment in offspring rats induced by prenatal stress and to explore its mechanism involving the expression of insulin-like growth factor-2 (IGF-2) and activity-regulated cytoskeletal-associated protein (Arc) in the hippocampus of the offspring.@*METHODS@#Rats were selected to establish a chronic unpredictable mild stress (CUMS) model during pregnancy. Offspring were weaned on 21st day and housed under either standard or an enriched environment. The learning and memory ability were tested using Morris water maze and Y-maze. The expression of IGF-2 and Arc mRNA and protein were respectively measured by using RT-PCR and Western blotting.@*RESULTS@#There was an elevation in the plasma corticosterone level of rat model of maternal chronic stress during pregnancy. Maternal stress's offspring exposed to an enriched environment could decrease their plasma corticosterone level and improve their weight. The offspring of maternal stress during pregnancy exhibited abnormalities in Morris water maze and Y-maze, which were improved in an enriched environment. The expression of IGF-2, Arc mRNA, and protein in offspring of maternal stress during pregnancy was boosted and some relationships existed between these parameters after being exposed enriched environment.@*CONCLUSIONS@#The learning and memory impairment in offspring of prenatal stress can be rectified by the enriched environment, the mechanism of which is related to the decreasing plasma corticosterone and increasing hippocampal IGF-2 and Arc of offspring rats following maternal chronic stress during pregnancy.

ABSTRACT

This study aimed to determine and evaluate whether there are differences in values of MAC in wildlife captured guan (Penelope obscura) under different conditions of social stress. This study used 27 bird species guan (P. obscura), divided into two groups: animals kept in the same enclosure (Collective Group) and animals kept in individual cages (Individual Group). The research was conducted at the Advanced Research Base of IBAMA, Painel/SC, and at the Clinical Veterinary Hospital of the "Universidade do Estado de Santa Catarina", Lages/SC. The birds were captured using network trap with manual disarmament and transported to the Veterinary Hospital in cages. The animals were fasted for 2 to 6 hours before the procedure; anesthetic induction was performed with isoflurane for instrumentation. The isoflurane CAM was placed in the target value 1.3v% in the first animal of each group, and waited 15 minutes for the nociceptive (electric) stimulus, in the value of 50 hertz and 50mA, held in faradic form (3 consecutive simple stimuli, followed by 2 continuous stimuli). The stimulus was performed on the lateral side of the left pelvic limb in the tibiotarsal region, and the electrodes were fixed with a 22G needle at a distance of 5cm between them. The bird's responses to the stimulus were considered positive (wing movements, head or vocalization) or negative (not presented movements of wings, head or vocalization) and the MAC value of the animal was recorded. Negative responses reduced next birds' MAC value by about 10%, and MAC positive responses increased by about 10%. Statistical analysis was done by methods up and down and analyze quantal for MAC and paired t-test for equivalent or t-test for variances not equal variances for the physiological variables. At the end of the experiment, the birds were reintroduced in the same capture area. The MAC value of isoflurane in the Collective Group was 1.4v% and the Individual Group 1.9v% to 0.903atm. It is observed that guan (P. obscura) in the Collective Group showed lesser anesthetic resistance to isoflurane than the birds in the Individual Group, showing that some levels of social stress can influence the MAC values of the isoflurane.(AU)


Este estudo teve como objetivo determinar e avaliar se há diferença nos valores de CAM em jacus (Penelope obscura) capturados em vida livre e submetidos a diferentes condições de estresse social. Foram utilizadas 27 aves da espécie jacu (P. obscura) de vida livre, que depois de capturados foram alocados em dois grupos: 10 animais que permaneceram em grupo no mesmo recinto (Grupo Coletivo) e 17 animais que permaneceram em gaiolas individuais (Grupo Individual). A pesquisa foi realizada na Base de Pesquisa Avançada do IBAMA, Painel/SC, e no Hospital de Clínica Veterinária da Universidade do Estado de Santa Catarina, Lages/SC. A captura foi realizada utilizando armadilha de rede com desarmamento manual e as aves foram transportadas para o Hospital Veterinário em gaiolas. Os animais foram submetidos a um jejum alimentar de 2 a 6 horas antes do procedimento, a indução anestésica foi realizada com isoflurano para instrumentação. A CAM de isoflurano foi colocada no valor alvo 1,3v% no primeiro animal de cada grupo, e esperado 15 minutos para realização do estímulo nociceptivo (elétrico), no valor de 50 hertz e 50mA, realizado de forma farádica (3 estímulos simples consecutivos, seguidos de 2 estímulos contínuos). O estímulo foi realizado na face lateral do membro pélvico esquerdo na região tibiotársica, e os eletrodos fixados com agulha 22G a uma distância de 5cm entre elas. A resposta da ave ao estímulo foi considerada positiva (movimentos de asas, cabeça ou vocalização) ou negativa (não apresentou movimentos de asas, cabeça ou vocalização) e o valor de CAM do animal foi registrado. Para resultados negativos, a CAM da próxima ave foi reduzida em torno de 10%, para positivos a CAM foi aumentada em torno de 10%. A análise estatística foi feita pelos métodos up and down e análise quantal para a CAM e teste t de pareado para variâncias equivalentes ou teste t para variâncias não equivalentes para as variáveis fisiológicas. Ao final do experimento as aves utilizadas foram reintroduzidas na mesma área de captura. O valor da CAM de isoflurano no Grupo Coletivo foi de 1,4v% e no Grupo Individual a CAM de 1,9v% a 0,903atm, sendo o valor do Grupo Coletivo significativamente menor que o Grupo Individual. Observa-se assim que os jacus (P. obscura) que permaneceram em recinto coletivo apresentaram uma menor resistência anestésica ao isoflurano que as aves que permaneceram em recintos individuais, mostrando que alguns níveis de estresse social como os observados aqui podem influenciar sobre os valores da CAM do isoflurano.(AU)

ABSTRACT

Objective To evaluate the clinical efficacy of Yangxue-Pinggan decoction on intractable headache. Methods A total of 66 patients with intractable headache who met the inclusion criteria were randomly divided into two groups by random number table method, 33 in each group. The control group was given flunarizine hydrochloride capsule orally, while the treatment group was given professor Guan Youbo's Yangxue-Pinggan decoction. Both groups were treated for 4 weeks. The improvement rates of headache were observed. The headache attacks, the degree of headache, the duration of headache and the accompanying symptoms were scored, and the clinical efficacy was evaluated. Results The total effective rate was 97.0% (32/33) in the treatment group and 63.6% (21/33) in the control group, which there was significant difference between the two groups (χ2=11.591, P=0.001). After treatment, the scores of the headache attacks, the degree of headache, the duration of headache and the accompanying symptoms in the treatment group were significantly lower than those in the control group (t=17.689, 12.523, 26.907, 25.569, P<0.001). Conclusions The Yangxue-Pinggan decoction can obviously improve the clinical symptoms of intractable headache, relieve the pain degree and reduce the number of attacks, and its clinical efficacy is better than oral flunarizine hydrochloride capsule.

Se presenta el caso clínico de una lactante de 8 meses de edad, de procedencia rural, quien ingresa en el Hospital Docente Infantil Norte de Santiago de Cuba por presentar fiebre, vómitos y diarreas. Se le realizó examen físico y observación de la vena digital, cuyo resultado desde el punto de vista de la medicina tradicional fue diarrea por frío-humedad, para lo cual se orientó una estrategia terapéutica que favoreció la mejoría del cuadro diarreico y emético, sin el empleo de medicamentos


The case report of an 8 months infant is presented. She had rural origin and is admitted to the Northern Teaching Children Hospital in Santiago de Cuba due to fever, vomits and diarrheas. Physical examination and observation of the digital vein was carried out whose result from the traditional medicine point of view was diarrhea due to cold-humidity, for which a therapeutic strategy was guided that favored the improvement of the diarrheal and emetic pattern, without using medication

ABSTRACT

This study aimed at efficiently screening out the toxic herbal medicines in Zhuang Gu Guan Jie pill (ZGGJ) based on the zebrafish model.Zebrafish embryos at 1 day post fertilization (dpf) were administered by ZGGJ and the water decoction (WD) and ethanol extract (EE) of 12 herbal drugs in it with various concentrations.The number of embryo deaths after the administration was recorded at 2-6 dpf and 2-8 dpf,respectively.Embryonic micro-morphology of zebrafish (3 dpf) was observed,LC50 value at 6 dpf was calculated by SPSS16.0.As a result,both the WD and EE of ZGGJ,fructus psoraleae,radix dipsaci,olibanum,myrrh,radix angelicae pubescentis and aucklandiae radix,and the EE of Caulis Spatholobo and Rhizoma Cibotii presented high organ toxicity to the juvenile zebrafish,featuring yolk sac swelling,deforming,blackening,indicating hepatotoxicity in the zebrafish.Their LC50 value at 6 dpf was from 33.2 μg·mL-1 to 392.7 μg·mL-1 (LD50 value was converted to crude drug.The same bellow.).The WD and EE of Epimedii Folium at 1,500 μg· mL-1 led to yolk sac swelling and blackening.The LC50 at 6 dpf varied at 1,100-1,350 μg·mL-1,showing a certain toxicity of Epimedii Folium.The WD of Caulis Spatholobo and Cibotii Rhizoma and both WD and EE of Rhizoma drynariae,taxilli herba and prepared rehmanniae radix didn' t change the toxicity to the organs in zebrafish.LC50 value were over 1,500 μg·mL-1,presenting favorable safety.In conclusion,it was demonstrated that the zebrafish model efficientlyidentified the toxicity of different herbal medicines from ZGGJ and efficiently screened out the potential toxicants in vivo with the advantages of real-time,dynamic and large scale for traditional Chinese medical compounds.

ABSTRACT

Professor GUAN Zun-xin is a national famous TCM doctor who enjoys the State Council Special Allowance. He has been engaged in clinical research on kidney diseases for a long time. In this article, Professor GUAN Zun-xin’s experience in the treatment of chronic renal failure was introduced. The combined therapy is named“GUAN’s five therapy for renal diseases”, including five therapies:cross medicine moxibustion, acupoint injection, earpins, traditional Chinese medicine herbs and happy care therapy. This therapy applies acupuncture, moxibustion and medicine at the same time, and pays attention to mental problems and physical problems. Acupuncture is used to smoothen meridians, adjust balance, and promote functions. Medicine is used to reinforcing deficiency and reducing excess, with a purpose to provide references for other TCM doctors.

ABSTRACT

This study was aimed to predict active compounds,drug targets and potential diseases of Yi-Guan decoction (YGD) by network pharmacological technology,and to clarify molecular mechanisms of YGD efficiency on different diseases with liver and kidney yin deficiency syndrome (LKYDS).Chemistry compounds,targets and related diseases from YGD were collected from TCMSP,TCM Database@Taiwan,TCMID,HIT,Drugbank,PubChem and TTD databases.The YGD compounds-targets-diseases network was constructed.The network topology was analyzed by Cytoscape software.The analysis of GO biological processes and KEGG pathways enrichment were performed by DAVID website.The results showed that 849 chemical compounds were identified from Beishashen,Maidong,Danggui,Shengdihuang,Chuanlianzi and Gouqizi.There were 49 active CHM compounds that were both oral bioavailability (OB) ≥ 30% and drug-likeness (DL) ≥ 0.18,corresponding to 200 target proteins and 264 diseases.The top three GO biological processes were response to organic substance,regulation of cell proliferation,and response to endogenous stimulus,respectively.The top three KEGG pathways were pathways in cancer,hepatitis B,and prostate cancer,respectively.It was concluded that the analysis on YGD was conducted based on network pharmacology,and the compound-target-disease network was built,which may help to clarify the mechanisms of YGD efficiency on different diseases with LKYDS.It can provide clues to find new potential clinical adaptation of disease and new drugs.

[Objective] To explore the location of Renying and Qikou pulses, and its clinical role. [Method] Through studying the relevant contents of the Renying Qikou pulse in <Huangdi Neijing>, and accessing to the relevant information, combining in with the clinical experience of the author, to analyze the location and clinical function of the pulse. [Result] There are plenty of Renying and Qikou pulse in<Huangdi Neijing>,but most of scholars think that the Renying is located in the neck since ancient times, which is difficult to use in clinical practice. The author thinks that the Renying is located in a point ahead of the left Guan Pulse, and Qikou is located in a point ahead of the right, and introduces the method to judge size, through the author 's literature reviews and clinical experience, for the efficient diagnosis and treatment. [Conclusion]The Renying Qikou pulse is one of the most main contents in<Huangdi Neijing>, which has rich information. The pulse has an effect that can not be ignored for modern Chinese medicine clinic, especially for the acupuncture and moxibustion.

ABSTRACT

In the field of traditional Chinese medicine(TCM),the definition of "Shen" contains a broad concept and a narrow concept. Broad "Shen" is the dominator and administrator of human vital movement,while narrow "Shen" means spirit, consciousness, thinking and emotions of human. "Shen" originates from innate essence of parents, and survives on acquired essence of water and food as well as the nutrition of its metaplastic Qi-Blood-Body fluid. Qi-Blood,the most precious asset of human body, is a quite important section of one person and the central material basis of "Shen-Zhi". Coronary heart disease(CHD) is classified as thoracic obstruction category in TCM. However, in the progress of CHD, the mental factors and physical factors interplay each other. Thus, CHD is defined as a kind of psychosomatic disorder. So, the concept of psycho-cardiology generally corresponds to the cognition of TCM. Disorder of Qi and Blood, Qi deficiency and Blood stasis and lack of spirit and "Shen" preservation, are the main causes of coronary heart disease(CHD). Guan-Xin-Jing capsule possesses multiple therapeutical effects including supplementing qi and activating blood circulation, tranquilizing "Shen" and sedating "Zhi" and balancing psychosomatic status. It will be of vital importance and promotional value in the prophylaxis and treatment of CHD.

ABSTRACT

Objective To observe the clinical efficacy of pestle acupuncture at Yao Yang Guan Ba Zhen and He Che Ming Qiang Duan in treating lumbago due to cold and damp. Methods Totally sixty-two patients with lumbago due to cold and damp were randomized into a treatment group and a control group, 31 in each group. The two groups were both intervened by medicinal fumigation, while the treatment group was additionally treated with pestle acupuncture at Yao Yang Guan Ba Zhen and He Che Ming Qiang Duan and the control group was additionally with ordinary acupuncture. Visual Analogue Scale (VAS) was observed before and after intervention, and the clinical efficacies were compared. Results The total effective rate was 90.3%in the treatment group versus 87.1% in the control group, and the difference was statistically significant (P<0.05). The VAS scores were significantly changed after intervention in both groups (P<0.01). After intervention, there was a significant difference in comparing VAS score between the two groups (P<0.05). Conclusions Pestle acupuncture is an effective method in treating lumbago due to cold and damp, and it doesn’t cause any injury or pain.

This study was aimed to discuss the difference of Cun-Kou six-pulse (CKSP) diagram of different diseases. The ZBOX-I Type Pulse Digital Analyzer was used to collect CKSP diagram of 30 bronchial asthma cases and 33 chronic gastritis cases. Time-domain parameter was extracted for analysis. Comparison was made by statistical methods. The results showed that there was statistical difference on time-domain parameter of pulse diagram for different diseases (P<0.05). There was statistical difference on time-domain parameter of pulse diagram between the left side and right side (P<0.05). There were statistical differences on t4 of left Cun, t4 and h5 of right Cun, t1\t4 of left Chi between bronchial asthma and chronic gastritis cases (P<0.05). It was concluded that the single-cycle pulse diagram of chronic gastritis was wider but no higher than that of bronchial asthma. The systole phase of single-cycle pulse diagram of bronchial asthma occupied greater proportion than that of chronic gastritis. There was difference on Cun of time-domain parameter in pulse diagram between bronchial asthma and chronic gastritis. It was consistent with the theory of Cun-Kou sub-designated zang-fu organs, which was “left Cun designates heart, right Cun designates lung.” However, there was no further explanation on functional differences between the spleen and stomach. The mechanism of pulse difference on the left and right side still required further illustration.

ObjectiveTo observe the effect ofTong Guan Li Qiao(opening orifices) needling method on the swallowing function in patients with post-stroke deglutition disorders.MethodNinety-one patients were randomized into an acupuncture group of 46 cases and a control group of 45 cases. The acupuncture group was intervened byTong Guan Li Qiaoneedling method, while the control group was by rehabilitation training. The Standard Swallowing Assessment (SSA), Modified Barthel Index (MBI), Swallowing-related Quality of Life (SWAL-QOL) and Hamilton Depression Scale (HAMD) were adopted for evaluation before and after intervention.ResultAfter intervention, the SSA, MBI, SWAL-QOL, and HAMD scores were significantly improved in the two groups (P<0.05); there were significant differences in comparing the scores between the two groups after intervention (P<0.05), and the improvements in the acupuncture group were more significant than that in the control group (P<0.05).Conclusion Tong Guan Li Qiaoneedling method can produce a more significant efficacy than conventional rehabilitation, by more effectively improving the swallowing function, activities of daily life, quality of life, and depression state in patients with post-stroke deglutition disorders.

ABSTRACT

[Objective] Research for the pulse mechanism, diagnosis and treatment of Pro. Lian Jianwei on right guan large virtual pulse. [Method] Through analyzing Pro. Lian learning process of pulses and his outpatient cases, explore his clinical diagnosis and treatment on right guan large virtual pulse, summarize the right guan large virtual pulse mechanism and the use of formula and medication by Pro. Lian. [Result] Pro. Lian attaches importance to the value of Li Dongyuan’s academy of spleen and stomach, skil in pulse diagnosis. He considers that right guan large virtual pulse is the pulse on right hand guan which is wide and large, soft and empty when taking pulse. Internal injury due to emotional disorder and diet fatigue are the main factors that result to deficiency of spleen qi and stomach qi, endophytic of yin-fire, consumed yin-blood, causing qi detached from blood and lead to the rising of qi to surface. The treatment obeys the rules of“debilitated patients treated with herbs warm in property ”by“Neijing”, uses Li Dongyuan’s Buzhongyiqi decoction with herbs sweet in flavor and warm in property to invigorate qi to elevate spleen-yang, for sufficient primordial qi and convergence yin-fire, also modifying the formula accord-ing to the patient’s syndrome. [Conclusion] Pro. Lian is good at inheriting Li Dongyuan’s spleen and stomach academy, attaches importance to the pulse diag-nosis in his clinical practice, good at using Li Dongyuan’s Buzhongyiqi decoction and modifying it according to the patient’s syndrome as the treatment for right guan large virtual pulse.

ABSTRACT

This study was performed to investigate the effect of Nei Guan (PC-6) moxibustion stimulation on artificial bradycardia of dogs. Xylazine was injected for inducing bradycardia. Rectal temperature, systolic blood pressure, respiratory rate, heart rate were recorded every 10 minutes for 120 minutes. Systolic blood pressure significantly increased on 40 min (p < 0.05) after xylazine injection, compared with those of control group. Heart rate significantly increased on 40 min (p < 0.01), 50 min (p < 0.01), 60 min (p < 0.01), 70 min (p < 0.01), 80 min (p < 0.01), 100 min (p < 0.01), 120min (p < 0.01) after xylazine injection, compared with those of control group. In conclusion, moxibustion of Nei Guan (PC-6) showed recovery effect in xylazine induced bradycardia in dogs.

ABSTRACT

[Objective] To investigate the effect of electroacupuncture (EA) of Neiguan (PC6) poinl on rnyocardial fibrosis and myocardial contents of collagen type Ⅰ and type Ⅲ in rats with pressure overload. [ Methods ] Rats were randomized to four groups: pseudo-operation group ( group A), model group ( group B) , EA group ( group C) and western medicine group (group D). Pressure-overloaded rat models were established by restriction of abdominal aortic artery. Left ventricular mass index (LVMI) and blood pressure (BP) in the four groups were observed; the plasma level of angiotensin Ⅱ (Ang Ⅱ ) was detected by radioimmunoassay and myocardial contents of collagen type Ⅰ and type Ⅲ by immunohistochemical staining method. [Results] BP, LVMI, expression levels of Ang If and collagen type Ⅰ and type Ⅲ were increased in group B as compared with those in group A ( P 0.05) . [Conclusion] EA of Neiguan point is effective in preventing and treating myocardial fibrosis induced by pressure overload.
